General description

Novobiocin sodium salt is an antibiotic derived from soil bacteria. Novobiocin inhibits the action of bacterial DNA gyrase and belongs to a class of compounds called coumarin antibiotics. Coumarins are heterocyclic compounds with a wide range of biological activity, including antibacterial, anticoagulant, antioxidant, anti-inflammatory, antiviral, antitumor, and enzyme inhibitory effects.

Application

For the production of positively supercoiled plasmid DNA. Inhibitor of bacterial DNA gyrase and eukaryotic DNA topoisomerase. Inhibitor of retrovirus RNA-dependent DNA-polymerase.

Novobiocin Sodium Salt, an inhibitor of Hsp90, can be used:


  • in the research heat shock protein inhibition and to produce positively supercoiled plasmid DNA[1]
  • in the research on the efficacy of probiotics[2][3]
  • in the research on the efficacy of human monocytes and dendritic cells for T-cell replication[4]

Biochem/physiol Actions

Mode of Action: Inhibits DNA synthesis by inhibiting the enzyme Topoisomerase II.

Antimicrobial spectrum: Gram-positive bacterial.

Novobiocin inhibits DNA synthesis by inhibiting the GyrB subunit of the Topoisomerase enzyme involved in energy transduction. Novobiocin acts as a competitive inhibitor of the ATPase reaction catalysed by GyrB[1].
